Product Specific Information

The ICRF44 monoclonal antibody reacts with human CD11b, a 165 kDa adhesion molecule. CD11b, associated with integrin β2 (CD18), is expressed on monocytes, granulocytes, activated lymphocytes, and a subset of NK cells. It serves as a receptor for ICAM family members (CD54, CD102, CD50) and iC3b, playing a key role in cell-cell and cell-matrix interactions.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells at 5 µL (0.25 µg)/test.

Super Bright 600 is a tandem dye excited by the violet laser (405 nm) and emits at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 2–8°C in the dark with minimal impact on brightness.

Target Information

CD11b (integrin alpha-M, ITGAM, integrin alpha-X, ITGAX) is a 165 kDa adhesion molecule forming a heterodimer with integrin beta-2 (CD18), known as Mac-1 or CR3. It is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, NK cells, dendritic cells, and microglia. CD11b/CD18 mediates adhesion via ICAMs, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X—essential for cell-cell and cell-matrix interactions.

Expression patterns:

• 8% of spleen cells
• 44% of bone marrow cells
• <1% of thymocytes

Commonly used as a microglial marker in nervous tissue. Expression increases during monocyte maturation and varies among macrophage subsets. Dysregulation of CD11b is associated with systemic lupus erythematosus and ITGAM-related susceptibility.
