Target Information

The expression pattern of the Prox1 homeobox gene suggests that it plays a role in various embryonic tissues, including the lens. Analysis of mRNA reveals that Prox1 is present in many human tissues, with the lens showing the highest level. Homozygous Prox1- mice die at midgestation due to multiple developmental defects, including lens abnormalities. Prox1 inactivation leads to abnormal cellular proliferation, downregulation of Cdkn1b and Cdkn1c, misexpression of E-cadherin, and excessive apoptosis. Consequently, mutant lens cells fail to polarize and elongate properly, resulting in a hollow lens.

Prox1 is expressed in a subpopulation of endothelial cells that form the lymphatic system through budding and sprouting. It is a specific and essential regulator of lymphatic system development. Prox1 is also required for hepatocyte migration in mice; its loss results in smaller livers with fewer clustered hepatocytes. Additionally, Prox1 regulates progenitor cell egress from the cell cycle in the embryonic retina — loss of Prox1 reduces cell cycle exit, while ectopic expression promotes it. Overall, Prox1 is a key regulator of progenitor-cell proliferation and cell-fate determination in the vertebrate retina.
