Target Information

Isoform 1 functions as an extracellular chaperone that prevents aggregation of nonnative proteins and stress-induced aggregation of plasma proteins. It inhibits amyloid fibril formation by various proteins (APP, APOC2, B2M, CALCA, CSN3, SNCA, etc.) and maintains partially unfolded proteins for subsequent refolding by other chaperones such as HSPA8/HSC70.
It does not refold proteins by itself but mediates internalization and degradation of chaperone-client complexes. Secreted isoform 1 protects cells against apoptosis and complement-mediated cytolysis. Intracellular isoforms interact with ubiquitin and SCF E3 ligase complexes to promote proteasomal degradation of target proteins and modulate NF-κB activity.
Nuclear isoforms promote apoptosis, while mitochondrial isoforms suppress BAX-dependent cytochrome c release and inhibit apoptosis. Plays a role in regulating cell proliferation.
