Target Information
Sonic Hedgehog (SHH) is expressed only during embryogenesis and plays a crucial role in early embryonic patterning. It is a key inductive signal in the formation of the ventral neural tube, anterior-posterior limb axis, and ventral somites. The SHH precursor undergoes autocatalytic cleavage, generating an N-terminal signaling domain and a C-terminal processing domain. The C-terminal product attaches a cholesterol moiety to the N-terminal product, restricting its diffusion. Defects in SHH or its signaling pathway can cause holoprosencephaly (HPE), characterized by abnormal forebrain development and facial deformities, and may also be linked to VACTERL syndrome involving multiple congenital anomalies.
