Target Information
Apoptosis, or programmed cell death, plays major roles in development and normal tissue turnover, as well as tumor formation. During apoptosis, gene expression patterns change significantly. One such gene, PDCD4 (Programmed Cell Death Protein 4), is upregulated following apoptosis onset. PDCD4 encodes a tumor suppressor protein whose expression is lost in breast, colon, lung, and prostate carcinomas. It binds and inhibits the helicase activity of eukaryotic translation initiation factor 4A and suppresses AP-1-mediated transactivation and transformation. Akt kinase regulates PDCD4 by phosphorylation, reducing its inhibitory effect on AP-1-responsive promoters. Two isoforms of PDCD4 are known.
