Target Information

TRPC6 forms a receptor-activated calcium channel in the cell membrane. The TRPC6 channel is activated by diacylglycerol (DAG) and is regulated by the phosphatidylinositol second messenger system. Activation occurs independently of protein kinase C and is not triggered by low intracellular calcium levels.
Defects in the TRPC6 gene are associated with focal segmental glomerulosclerosis 2 (FSGS2). TRPC6 belongs to the mammalian transient receptor potential (TRP) superfamily, specifically the “canonical TRP” (TRPC) family. Members of this family are activated through PLC-coupled receptors and act as receptor-regulated cation channels.
TRPC6/6/7 subfamily members can be activated by DAG analogs, suggesting a mechanism of activation via PLC-coupled receptors. When expressed in transfected cells, TRPC6 functions as a non-selective, store-independent receptor-activated cation channel. It is activated by DAG in a PKC-independent manner and is insensitive to IP3 activation. Increasing evidence indicates that TRPC6 encodes endogenous DAG-activated receptor-operated cation channels in vivo.
Diseases associated with TRPC6 include focal segmental glomerulosclerosis (FSGS2).
