Target Information
Smad4은 인간의 18번 염색체에 위치한 Smad4 유전자에 의해 암호화됩니다. Smad4는 Smad family member 4, Sma- and Mad-related protein 4, 또는 Mothers against decapentaplegic (MAD) homolog 4로도 알려져 있습니다.
이 단백질은 N-말단의 MH1 (MAD homology 1) 도메인과 C-말단의 MH2 (MAD homology 2) 도메인을 포함하며, 각각 DNA 결합과 단백질 상호작용에 관여합니다.
TGF-beta 및 BMP와 같은 TGF-beta 수퍼패밀리 리간드가 수용체에 결합하면 Smad1, 2, 3, 5, 8 (R-Smad)의 인산화가 일어나고, Smad4 (co-Smad)와 복합체를 형성하여 핵으로 이동합니다.
Smad6 및 Smad7과 같은 억제성 Smad (I-Smad)는 활성화된 R-Smad와 상호작용하여 신호 전달을 억제합니다.
Smad4는 종양 억제 단백질로 작용하며, Cyclin D1의 발현을 억제하고 collagen 발현을 촉진함으로써 세포 증식을 조절합니다. 탈인산화는 Smad의 핵 내외 이동을 조절합니다.
