Target Information
Smad4는 인간 염색체 18에 위치한 Smad4 유전자에 의해 암호화됩니다. Smad family member 4, Sma- and Mad-related protein 4, Mothers against decapentaplegic homolog 4 등으로도 알려져 있습니다.
Smad4는 N-말단의 MH1 도메인(DNA 결합)과 C-말단의 MH2 도메인(단백질 상호작용)을 포함합니다.
TGF-beta 또는 BMP 리간드가 수용체에 결합하면 R-Smad (Smad1, 2, 3, 5, 8)가 인산화되어 Smad4와 삼합체를 형성하고 핵으로 이동합니다.
Smad6, 7 등의 억제성 Smad는 활성화된 R-Smad와 상호작용하여 신호를 억제합니다.
Smad4는 Cyclin D1의 발현 억제 및 collagen 발현 증가를 통해 세포 증식을 억제하는 종양 억제 단백질로 작용합니다.
탈인산화는 Smad의 핵 수출 및 핵-세포질 간 이동을 조절합니다.
