Target Information
Smad4은 인간의 18번 염색체에 위치한 Smad4 유전자에 의해 암호화됩니다.
다른 이름으로는 Smad family member 4, Sma- and Mad-related protein 4, Mothers against decapentaplegic (MAD) homolog 4로 알려져 있습니다.
Smad4 단백질은 N-말단의 MH1 (MAD homology 1) 도메인과 C-말단의 MH2 (MAD homology 2) 도메인을 포함하며, 각각 DNA 결합과 단백질 상호작용에 관여합니다.
TGF-β 및 BMP와 같은 리간드가 수용체에 결합하면 Smad1, 2, 3, 5, 8 (R-Smad)이 인산화되고, Smad4 (co-Smad)와 복합체를 형성하여 핵으로 이동합니다.
Smad6 및 Smad7과 같은 억제 Smad(I-Smad)는 활성화된 R-Smad와 상호작용하여 신호를 억제합니다.
Smad4는 Cyclin D1의 발현 억제 및 collagen 발현 촉진을 통해 세포 증식을 억제하는 종양 억제 단백질로 작용합니다.
탈인산화는 Smad 단백질의 핵 내외 이동을 조절합니다.
