BACE1 (beta-secretase)는 Alzheimer’s disease(AD) 관련 아밀로이드 베타 펩타이드 생성에 관여하는 핵심 효소입니다. APP의 ‘Swedish’ 돌연변이는 BACE1에 의한 절단을 촉진하여 조기 발병형 AD를 유발할 수 있습니다. APP는 먼저 β-secretase에 의해 절단되어 용해성 단백질 파생물과 막 결합형 C99 단편을 생성하며, 이후 γ-secretase가 이를 절단하여 아밀로이드-β 펩타이드를 생성합니다. 이러한 경로는 AD 병리의 주요 원인으로, BACE1 및 관련 효소의 억제가 치료적 접근으로 연구되고 있습니다.
