The CoV Spike (S) protein plays a crucial role in viral attachment, fusion, and entry, serving as a key target for antibody, inhibitor, and vaccine development.
The receptor-binding domain (RBD) within the S1 subunit binds strongly to human and bat ACE2 receptors, exhibiting higher affinity than SARS-CoV RBD.
S1 contains the RBD responsible for ACE2 binding, while S2 mediates membrane fusion.
SARS-CoV-2 S protein consists of 1273 amino acids: a signal peptide (1–13), S1 subunit (14–685), and S2 subunit (686–1273). The extracellular domain (S-ECD) represents the Spike protein’s functional region.
