The CoV Spike (S) protein is crucial for viral attachment, fusion, and entry, serving as a major target for antibody and vaccine development.
The receptor-binding domain (RBD) of the Spike protein binds strongly to human and bat ACE2 receptors, exhibiting higher affinity to ACE2 than SARS-CoV RBD.
S1 contains the RBD responsible for ACE2 binding, while S2 mediates membrane fusion.
SARS-CoV-2 S protein is 1273 amino acids long, with a signal peptide (aa 1–13), S1 subunit (aa 14–685), and S2 subunit (aa 686–1273).
The S-ECD represents the extracellular domain of the Spike protein.
