DjC9 (DnaJ subfamily C member 9) belongs to the Hsp40 (heat shock protein 40) family, functioning as a co-chaperone to Hsp70.
Hsp40 proteins are classified into three subfamilies (A, B, and C), defined by the presence of specific conserved domains such as:
- J-domain (alpha-helical N-terminal domain)
- Glycine/phenylalanine-rich region
- Cysteine-rich region
- C-terminal beta-sheet domain
DjC9, part of subfamily C, contains only the J-domain and interacts with HSP70 through this domain, activating its ATPase activity.
