Product Specific Information

The 2B11 monoclonal antibody reacts with mouse and human CXCR4 (CD184), also known as Fusin, LESTR, or HUMSTR. CXCR4 is a G-protein-coupled chemokine receptor functioning as a coreceptor for X4 HIV-1 entry into CD4+ cells and plays a role in lymphocyte migration. Its ligand is SDF1, essential for granulocyte progenitor maintenance and B cell development.

Expression differs between species:

  • Mouse thymus: restricted to CD4+CD8+ cells
  • Mouse spleen: predominant on B lymphocytes

2B11 recognizes the amino terminus of CXCR4 and induces receptor internalization without blocking SDF1a binding.

Applications Reported: Flow cytometric analysis
Applications Tested: Flow cytometric analysis of mouse thymocytes at ≤ 0.25 µg/test (100 µL final volume). Optimal titration recommended.

Light Sensitivity: Tandem dye sensitive to photo-induced oxidation. Protect from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark with minimal brightness impact. Clone-specific performance should be verified empirically.

Excitation: 488–561 nm
Emission: 775 nm

Target Information

CXCR4 (Fusin, LESTR, HUMSTR) is a seven-transmembrane G-protein-coupled receptor acting as a co-receptor for HIV-1 and HIV-2 entry into CD4+ cells. It binds CXCL12/SDF-1, mediating calcium signaling and MAPK activation, and serves as a receptor for extracellular ubiquitin. CXCR4 is involved in hematopoiesis, cardiac and gastrointestinal vascular development, neuronal survival, and inflammatory response to LPS.

Antibodies to CXCR4 can block HIV fusion and infection. The amino-terminal domain and second extracellular loop serve as HIV binding sites.
