Target Information

RUNX2 is a nuclear protein belonging to the RUNX family of transcription factors, characterized by a Runt DNA-binding domain. It is essential for both membranous and endochondral bone formation. RUNX2 regulates osteoblastic differentiation and skeletal morphogenesis, acting as a scaffold for nucleic acids and regulatory factors involved in skeletal gene expression.

The protein binds DNA as a monomer or as part of a heterodimeric complex with higher affinity. It plays a key role in enhancing TGFBR1 expression in osteoblasts and cooperates with DLX5 or related factors to activate osteoblast-specific gene expression. Mutations in RUNX2 are associated with bone development disorders such as cleidocranial dysplasia (CCD).
