Product Specific Information
Description:
The 4B12 monoclonal antibody reacts with mouse CCR7 (CD197), a chemokine receptor for CCL19 and CCL21. CCR7 mediates lymphocyte migration to secondary lymphoid organs and plays roles in dendritic cell and B cell localization during immune responses. It is also a marker for central memory T cells (TCM) and is involved in thymocyte development.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes and thymocytes. Recommended ≤1 µg/test in 100 µL volume. Optimize titration for best performance.
Staining Protocol (4B12 Antibody)
- Prepare cell suspension and block FcγR with 5 µg/million cells anti-mouse CD16/32 for 15 min on ice.
- Without washing, add 1 µg/million cells 4B12 and incubate at 37°C (preferred) or 2–8°C for 30 min.
- Wash cells once with 3 mL Flow Cytometry Staining Buffer and decant.
- Add secondary antibody and incubate at 2–8°C for 30 min.
- Wash once more and analyze by flow cytometry.
Note:
- Staining at 37°C yields brighter signal and better resolution.
- Co-staining with CCL19-Fc may be hindered due to steric interference.
- 4B12 binding can block CCL19-Fc binding, but not vice versa.

Target Information
CCR7 is a G protein-coupled chemokine receptor induced by Epstein-Barr virus (EBV) and expressed in blood, bone marrow, lymph node, and intestine. It regulates migration of memory T cells, dendritic cell maturation, and lymphocyte trafficking to inflamed tissues. CCR7 ligands include CCL19 (ELC) and CCL21 (SLC), essential for immune cell chemotaxis.
