Target Information
Janus kinase 2 (JAK2)는 특정 사이토카인 수용체 신호 전달 경로에 관여하는 단백질 티로신 키나아제로, polycythemia vera에서 돌연변이가 발견됩니다. JAK2는 JH1 촉매 도메인을 포함하며, JAK의 활성화는 STAT 단백질의 인산화 및 핵 이동을 유도하여 다양한 유전자 전사를 조절합니다.
JAK2는 IFN-γ 수용체 복합체의 개시를 위해 필요하며, JAK1은 증폭 역할을 수행합니다. 일부 연구에서는 JAK2의 역할이 Tyk2 및 JAK3에 의해 대체될 수 있다고 제안합니다.
JAK2 변이는 Budd-Chiari 증후군, Polycythemia vera, Thrombocythemia 3, Myelofibrosis, Acute myelogenous leukemia를 유발할 수 있습니다.
