Target Information
Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are transported within the cell by approximately 300 kDa cation-independent mannose 6-phosphate receptors (CI-MPR). The biosynthetic cycle involves MPR-mediated transport of newly synthesized lysosomal enzymes from the trans-Golgi network to late endosomes or early lysosomes. The endocytic cycle moves extracellular lysosomal enzymes from the plasma membrane via clathrin-coated vesicles to early endosomes. MPRs cycle between these compartments every 3 hours, with the majority located in late endosomes, fewer in the trans-Golgi network, and about 10% at the cell surface. MPR also binds type II insulin-like growth factor (IGF-II) simultaneously with man6P, and a soluble truncated form (~240 kDa) representing its extracellular domain circulates in serum with dual ligand-binding capability.
