Target Information

Specialized cells at the midline, which separates the left and right halves of the central nervous system (CNS), play key roles in directing growth cone behavior. In the vertebrate spinal cord, insect ventral nerve cord, and C. elegans, midline cells produce guidance cues such as nectins and slit, acting as attractants and repellents, respectively.

Robo (Roundabout) functions as an axon guidance receptor, part of the immunoglobulin superfamily conserved from fruit flies to mammals. It serves as a receptor for the repellent Slit and operates in a cell-autonomous manner. Non-crossing axons express high levels of Robo, while crossing axons express low levels before reaching the midline and high levels after crossing.

Robo1 and Robo2 are human homologs of the Drosophila Roundabout protein. Robo1 is also homologous to the C. elegans gene sax3, and Robo2 to the zebrafish gene astray.
