Target Information

The munc-18 interacting protein (Mint) family comprises evolutionarily conserved adaptor proteins involved in membrane transport and organization. Mammals express three isoforms: Mint1, Mint2, and Mint3.
The C-terminal region of these proteins is highly conserved and contains phosphotyrosine-binding (PTB) and PSD-95/DLG-A/ZO-1 (PDZ) domains, which serve as protein interaction modules.

• Mint1 and Mint2: Expressed mainly in the brain; interact with Munc18, a synaptic vesicle fusion component.
• Mint3: Expressed ubiquitously with lowest levels in brain and testis; does not interact with Munc18.
  Instead, Mint3 interacts with the Alzheimer’s disease-related amyloid precursor protein (APP) through its PTB and PDZ domains.
  It is proposed that Mint3 links APP to transport machinery components, regulating its transport, endocytosis, and metabolism.
  Abnormal APP metabolism is associated with early-onset Alzheimer’s disease.
