Target Information
Smoothened drosophila homolog (SMOH) is a serpentine protein belonging to the G-protein coupled receptor Fz/Smo family with 787 amino acids. It has seven transmembrane domains, four glycosylation sites, and a putative extracellular amino terminus.
SMOH is activated by β-Arrestin 2 and GRK2, acts as a receptor for Sonic hedgehog (Shh), and serves as a positive regulator of the hedgehog signaling pathway along with patched protein (PTCH). It regulates vertebrate developmental processes, cellular proliferation, and differentiation by activating transcription factor Gli.
Mutations in Smoothened are associated with sporadic basal cell carcinoma of the skin. Expression is observed in heart and brain tissues.
