Target Information

The DDX3 protein is a member of the DEAD-box protein family, characterized by the conserved Asp-Glu-Ala-Asp (DEAD) motif and ATP-dependent RNA helicase activity. It exhibits high levels of RNA-independent ATPase activity, and unlike most DEAD-box helicases, its ATPase activity is stimulated by both RNA and DNA.

DDX3 plays roles in both the nucleus and cytoplasm:

  • Nuclear functions: transcriptional regulation, mRNP assembly, pre-mRNA splicing, and mRNA export
  • Cytoplasmic functions: translation, cellular signaling, and viral replication

Misregulation of DDX3 has been implicated in tumorigenesis. The gene has a paralog on the Y chromosome (DDX3Y), and pseudogenes with similarity to both DDX3 and DDX3Y are found on chromosome 4 and the X chromosome. Alternative splicing generates multiple transcript variants.
