Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is prostate carcinoma.
Target Information
FOXO1 (FKHR, Forkhead Box O1) is a transcription factor belonging to the forkhead homeotic gene family.
It can act as a coactivator or corepressor of nuclear receptor activity through the LXXLL motif in its C-terminal region.
FOXO1 association with PAX3 has been implicated in alveolar rhabdomyosarcoma.
Studies link FOXO1’s anti-tumor activity, autophagy, and myogenic differentiation.
It is the main target of insulin signaling regulating metabolic homeostasis under oxidative stress.
FOXO1 binds to insulin response elements (IRE) and Daf-16 family binding elements (DBE), controlling redox balance, osteoblast numbers, and bone mass.
It also regulates skeletal endocrine function and glucose metabolism by suppressing RUNX2 activity.
In hepatocytes, FOXO1 promotes gluconeogenesis with PPARGC1A, activating genes such as IGFBP1, G6PC, and PPCK1.
FOXO1 is a key regulator of cell death downstream of CDK1, PKB/AKT1, and SKT4/MST1.
