Target Information
CXCR4 (also known as Fusin, LESTR, HUMSTR) is a member of the G-protein-coupled chemokine receptor family with seven membrane-spanning domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and also for HIV-2 binding via gp120, promoting viral fusion. CXCR4 serves as the receptor for the chemokine CXCL12/SDF-1, mediating calcium signaling and MAPK activation. It also acts as a receptor for extracellular ubiquitin, influencing intracellular calcium and cAMP levels. CXCR4 is involved in hematopoiesis, cardiac and vascular development, and neuronal survival. It mediates inflammatory responses through LPS binding and TNF secretion by monocytes. Antibodies against CXCR4 can block HIV-1 and HIV-2 fusion and infection of human cells.
