Target Information
IKK alpha/beta is encoded by CHUK (conserved helix-loop-helix ubiquitous kinase) and IKKB (inhibitor of kappa light polypeptide gene enhancer in B-cells) located on chromosomes 10 and 8, respectively, in humans.
Upon stimulation by agents that activate the NF-κB pathway such as TNF alpha, the IKK complex subunits (IKK alpha and IKK beta) along with the regulator NEMO (IKK gamma) phosphorylate IκB proteins. This leads to ubiquitin-mediated degradation and activation of NF-κB, resulting in nuclear translocation.
Phosphorylation at serine 176 and 180 in the activation loop induces a conformational change required for kinase activation.
