  • For reconstitution, add 100 µL distilled water to achieve ~1 mg/mL concentration.
  • For conjugation experiments, perform additional desalting using Zeba Spin Desalting Columns (7K MWCO, 0.5 mL, Product #89882).

Target Information

Retinoic acid-inducible gene I (RIG-I) is a pattern recognition receptor (PRR) that detects viral double-stranded RNA (dsRNA) and activates innate immune responses.
RIG-I and MDA5 are RNA helicases with overlapping and distinct functions in viral recognition. RIG-I detects short dsRNA lacking a 5’-triphosphate end, while MDA5 recognizes long dsRNA.
Upon activation, both signal through IPS-1, leading to activation of NF-κB and IRF-3, and ultimately apoptosis, cytokine signaling, and inflammation.
RIG-I plays a key role in signaling against influenza A/B, RSV, paramyxoviruses, Japanese encephalitis virus, and West Nile virus.
MicroRNA-146a negatively regulates RIG-I targets (TRAF6, IRAK1, IRAK2), providing feedback inhibition.
Recent evidence also suggests RIG-I detects cytosolic DNA via RNA polymerase III activity.
