Product Specific Information

The TL2.1 monoclonal antibody reacts with human Toll-like receptor 2 (TLR2). TLR2 and TLR4 are pattern recognition receptors involved in innate immunity and inflammation. TL2.1 is a blocking antibody used to study TLR2-mediated cytokine production in human peripheral blood mononuclear cells (PBMC).
It immunoprecipitates human TLR2 (~90 kDa) from PBMC and HMEC.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (1 µg) per test (100 µL final volume). Cell count range: 10⁵–10⁸ cells/test.
Excitation: 488 nm; Emission: 520 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

TLR2 is a member of the Toll-like receptor family, essential for pathogen recognition and activation of innate immunity. It forms heterodimers with TLR1 or TLR6 to recognize bacterial and mycoplasma lipoproteins.
TLR2 is highly expressed in peripheral blood leukocytes and mediates host response to Gram-positive bacteria and yeast via NF-κB activation.
Expression is regulated by immune modifiers:

• Upregulation: LPS, GM-CSF, IL-1, IL-10
• Downregulation: IL-4, IFN-γ, TNF

TLR2 is expressed in tonsils, microglia, lymph nodes, appendices, and activated B-cells. CD14⁺ monocytes show the highest expression, followed by CD15⁺ granulocytes, CD19⁺ B-cells, and CD3⁺ T-cells.
