Target Information
Vinculin (VCL)은 세포-세포 및 세포-세포외기질 부착성 접합(adherens junction)과 관련된 세포골격 단백질입니다. 이 단백질은 F-actin을 세포막에 고정시키는 여러 상호작용 단백질 중 하나로 작용합니다. Vinculin은 integrin의 β 서브유닛이 talin에 결합하고, talin이 vinculin에 결합하며, vinculin이 alpha-actinin 및 자기 자신과 상호작용하는 일련의 분자적 상호작용을 통해 부착반(adhesion plaque)의 형성에 관여합니다. Alpha-actinin이 actin 필라멘트를 교차 결합하기 때문에, vinculin은 세포외기질과 세포질 미세필라멘트 시스템을 연결하는 핵심 요소로 작용합니다.
VCL 유전자의 돌연변이는 확장성 심근병증(dilated cardiomyopathy 1W) 또는 가족성 비후성 심근병증(familial hypertrophic 15)을 유발할 수 있습니다.
