Product Specific Information

The 236A/E7 antibody reacts with human Foxp3 protein (FORKHEAD BOX P3, SCURFIN, JM2). Foxp3 (49–55 kDa) is a transcriptional regulator of the forkhead/winged-helix family, identified as the gene defective in scurfy (sf) mice. High expression of Foxp3 mRNA is observed in CD4+CD25+ regulatory T cells (Treg), and ectopic expression in CD4+CD25− cells induces a Treg phenotype.

Intracellular staining and flow cytometric analysis of human PBMCs using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523) reveals staining of the CD4+CD25^bright population.

The epitope recognized by 236A/E7 differs from that of PCH101 (Product #72-5776-40). This antibody also recognizes rhesus macaque, sooty mangabey, and cynomolgus macaque.

Applications Reported: Intracellular staining, flow cytometry, and IHC.
Applications Tested: Pre-titrated and validated for intracellular staining and flow cytometry using the Foxp3/Transcription Factor Buffer Set (Product #00-5523-00).
Recommended usage: 5 µL (0.25 µg) per test in 100 µL final volume. Applicable for 10^5–10^8 cells/test.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is 381 amino acids long and expressed highly in CD25+CD4+ regulatory T cells, low in CD4+CD25− cells, and absent in CD4−CD8+ cells. FOXP3 acts as a master regulatory gene and a specific marker for Treg cells. Mutations cause scurfy phenotype in mice and IPEX syndrome (immune dysfunction, polyendocrinopathy, enteropathy, X-linked) in humans. Expression of FOXP3 in cancer cells is regulated by p53 in response to DNA damage.
