M700A targets IFN gamma in ELISA and Western blot applications and shows reactivity with human samples.
This antibody (clone 2G1) has been successfully paired as the coating antibody in a sandwich ELISA with detection antibody M701B (biotinylated conjugate of clone B133.5).
Typical dilutions for sandwich ELISA include:

• Coating: 1 µg/mL
• Detection: 0.125–0.25 µg/mL

The immunogen is recombinant human IFN gamma, and the antibody detects IFN gamma with a predicted molecular weight of approximately 16 kDa.

Endotoxin concentration ≤10 EU/mg (tested by LAL assay).
M700A (clone 2G1) and biotinylated M701B (clone B133.5) have been used in combination with recombinant IFNg protein SIFNG in ELISA applications.

Target Information

IFN gamma (Interferon gamma, Type II interferon) is a macrophage activation factor and immune interferon produced primarily by T-lymphocytes and natural killer cells.
It is a dimeric glycoprotein (~45 kDa) consisting of two 146 amino acid subunits.
On SDS-PAGE, IFN gamma appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.
The biological activity of IFN gamma is species-specific; human IFN gamma does not cross-react with mouse.

Functions include:

• Antiviral and tumor antiproliferative activity
• Induction of MHC class I and II
• Macrophage activation
• Enhancement of immunoglobulin secretion by B lymphocytes
• Cytokine regulation and synergistic action with other cytokines

Activation occurs via IFN gamma receptor I/II and JAK-STAT pathway.
Human IFN gamma shares ~40% sequence homology with mouse IFN gamma.
It is upregulated by IL2, FGF basic, EGF and downregulated by vitamin D3 or DMN.
Mutations in the IFN gamma gene are associated with aplastic anemia.
