Product Specific Information

The RA3-6B2 monoclonal antibody reacts with the exon A-restricted isoform of mouse CD45, a 220 kDa surface molecule. The CD45R/B220 epitope is mainly expressed by B cell lineage from early Pro-B to mature B cells, but also on certain activated T cells, LAK cells, NK progenitors, and lpr/lpr mutant mouse T cells.

Applications Reported:
Used for immunohistochemical staining (frozen and paraffin sections) and flow cytometric analysis.

Applications Tested:

• Flow Cytometry: ≤ 0.5 µg/test
• IHC (Frozen): ≤ 20 µg/mL
• IHC (Paraffin): ≤ 20 µg/mL (low pH antigen retrieval recommended)
  Antibody should be titrated for optimal assay performance.

Target Information

CD45R (PTPRC) belongs to the protein tyrosine phosphatase (PTP) family, which regulates cell growth, differentiation, mitotic cycle, and oncogenic transformation. CD45 is expressed in hematopoietic cells and is essential for T- and B-cell receptor signaling. It exists in multiple isoforms via alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. CD45 regulates Src-family kinases, contributes to lymphocyte development, antigen signaling, and integrin-mediated survival pathways. Dysfunction in CD45 is associated with Severe Combined Immunodeficiency and Hepatitis C.
