Target Information
HLA-C는 HLA class I heavy chain paralogue에 속하며, heavy chain과 light chain(beta-2 microglobulin)으로 구성된 이합체입니다. Heavy chain은 막에 고정되어 있으며, class I 분자는 면역계에서 중심적인 역할을 하여 endoplasmic reticulum lumen에서 유래한 펩타이드를 제시합니다. 거의 모든 세포에서 발현되며, 약 45 kDa의 분자량을 가집니다.
유전자는 8개의 엑손으로 구성되어 있으며, 각각의 엑손은 leader peptide, alpha1/alpha2 domain(펩타이드 결합 부위), alpha3 domain, transmembrane region, cytoplasmic tail 등을 암호화합니다. Exon 2와 3의 다형성은 각 class I 분자의 펩타이드 결합 특이성을 결정합니다. 이러한 다형성은 골수 및 신장이식 시 타이핑에 활용됩니다. 100개 이상의 HLA-C 대립유전자가 보고되어 있습니다.
HLA 및 MHC 항체는 면역펩타이드오믹스(Immunopeptidomics) 연구에서 중요한 역할을 하며, HPLC-MS/MS를 통해 신항원(neoantigen)의 동정 및 특성 분석을 가능하게 합니다.
