Target Information
Sterol regulatory element-binding proteins (SREBPs) are transcription factors belonging to the basic helix-loop-helix leucine zipper family. Three isoforms are identified in mammalian tissues with distinct structures and functions.
SREBP-1a and SREBP-1c (originally cloned as ADD1) are products of alternative promoter usage of the SREBP-1 gene, while SREBP-2 is transcribed from a separate gene.
SREBPs exist as 120 kDa inactive precursors in the endoplasmic reticulum membrane. Upon activation, they are cleaved in the Golgi, producing a mature 60–78 kDa transcriptionally active fragment.
In liver and adipose tissues, SREBPs regulate lipid and cholesterol metabolism by inducing transcription of related genes.
SREBP-1 primarily controls triglyceride synthesis, whereas SREBP-2 is associated with cholesterol synthesis.
