Target Information
STAT3 (Signal Transducer and Activator of Transcription 3)는 세포 내 신호전달 및 전사 조절에 관여하는 STAT 단백질 패밀리에 속합니다.
STAT3는 receptor-associated kinase에 의해 인산화되어 핵으로 이동하며, cytokine 및 growth factor 자극에 반응하여 전사 인자로 작용합니다.
Coactivator인 CREB-binding protein이 STAT3의 전사 활성화에 필요하며, Interferon-α, Interferon-γ, EGF, PDGF, IL-6 등에 의해 활성화됩니다.
Tyrosine 705의 JAK1, JAK2에 의한 인산화는 STAT3의 이합체 형성, 핵 이동, DNA 결합에 필수적입니다.
STAT3 유전자는 인간 17번 염색체 q-arm에 위치하며, IFN-α에 의해 활성화되지만 IFN-β에는 반응하지 않습니다.
STAT3는 c-Jun, CREB 등과 상호작용하며, 세포 성장 및 apoptosis 조절 등 다양한 생물학적 과정에 핵심적 역할을 합니다.
또한 Rac1 GTPase는 STAT3의 활성을 조절하며, PIAS3 단백질은 STAT3의 특이적 억제인자입니다.
세 가지 alternative splicing isoform이 존재하며, STAT3 유전자 결손 마우스는 배아 단계에서 치명적인 것으로 알려져 있습니다.
