Target Information
Telomerase is a ribonucleoprotein polymerase that maintains telomere ends by adding the telomere repeat sequence TTAGGG. The enzyme contains a protein component with reverse transcriptase activity (encoded by the TERT gene) and an RNA component serving as a template for the repeat.
Telomerase expression is typically repressed in postnatal somatic cells, leading to progressive telomere shortening and cellular senescence. Dysregulation of telomerase in somatic cells may contribute to oncogenesis.
Studies in mice suggest telomerase may also aid in chromosomal repair, as de novo synthesis of telomere repeats can occur at double-stranded breaks. Multiple alternatively spliced TERT variants exist, some of which are not fully characterized. Alternative splicing is thought to regulate telomerase activity.
