Target Information
Caspase 8 (CASP8)는 cysteine-aspartic acid protease (caspase) 계열에 속하며, 세포 사멸(apoptosis)의 실행 단계에서 중심적인 역할을 합니다. Caspase는 비활성 전구체 형태로 존재하며, 내부의 보존된 aspartic 잔기에서 절단되어 활성형 효소로 전환됩니다. Caspase 8은 Fas 및 다양한 세포 사멸 자극에 의해 유도되는 programmed cell death에 관여합니다. N-말단의 FADD-like death effector domain(DED)을 통해 FADD 단백질과 상호작용하며, 활성화된 Caspase 8은 Caspase 9 등 하위 caspase를 활성화시켜 세포 사멸을 유도합니다. 또한 Caspase 8은 Huntington 병 환자의 뇌 조직 불용성 분획에서 검출되어 신경퇴행성 질환과의 연관성이 제시되었습니다. 다양한 스플라이싱 변이체가 보고되어 있으나, 모든 변이체의 full-length 서열은 확인되지 않았습니다.
