Product Specific Information
The BM8 monoclonal antibody reacts with mouse F4/80 antigen, an approximately 160 kDa surface receptor belonging to the EGF-TM7 protein family. The F4/80 antigen is a well-established marker for mature macrophages, though it may also be expressed by peritoneal eosinophils, Langerhans cells, and certain dendritic cell subsets. Expression begins early in myeloid development and can be upregulated by M-CSF stimulation.

- Applications Tested: Tested by flow cytometric analysis of mouse resident peritoneal exudate cells. Recommended ≤1.0 µg/test (100 µL final volume).

Super Bright 702 is a tandem dye excited with the violet laser (405 nm) and emits at 702 nm. Use a 710/50 bandpass filter. When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
Light Sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information
F4/80 antigen is a 160 kDa cell surface glycoprotein, a member of the EGF-TM7 family, sharing 68% amino acid identity with human EMR1. Expression varies during macrophage maturation and activation. It is expressed on macrophages in tissues such as liver (Kupffer cells), skin (Langerhans), brain (microglia), gut lamina propria, peritoneal cavity, lung, thymus, bone marrow stroma, and spleen red pulp. It may also appear on certain dendritic cells but is absent from macrophages in T cell zones of lymphoid organs. F4/80 may play a role in the generation of efferent CD8⁺ regulatory T cells.
