Target Information
Hereditary multiple exostoses (HME) is an autosomal dominant disorder characterized by the formation of cartilage-capped bony protuberances mainly located on long bones. EXT1 and EXT2 proteins form homo/heteromeric complexes in vivo, leading to their accumulation in the Golgi apparatus. Both are endoplasmic reticulum-localized type II transmembrane glycoproteins with glycosyltransferase activities involved in heparan sulfate (HS) polymerization. EXT2 provides D-glucuronyl (GlcA) and N-acetyl-D-glucosaminyl (GlcNAc) transferase activities, while EXT1 rescues defective HS biosynthesis and elevates transferase levels in mutated cells.
