Target Information
HMGB1 (High-mobility group box-1) protein은 핵 내 비히스톤 DNA 결합 단백질로 처음 보고되었으며, 손상된 세포에서 방출되어 염증성 사이토카인으로 작용합니다.
Mouse HMGB1은 215개의 아미노산으로 구성된 단일 사슬 폴리펩타이드로, 두 개의 DNA 결합 도메인(HMG box A, aa 9–79; box B, aa 89–162)과 음전하를 띤 C-말단 산성 꼬리 영역(30 aa)을 포함합니다.
Residues 28–44 및 180–185는 핵 위치 신호(NLS)를 포함하며, B box는 사이토카인 활성을, A box는 전사 인자의 helix-loop-helix 도메인과 연관됩니다.
HMGB1은 염증 매개체로 작용하여 단핵구 이동 및 사이토카인 분비를 유도하며, RAGE, TLR2, TLR4 수용체를 통해 신호 전달을 수행합니다.
모든 세포 핵과 세포질에서 보존적으로 발현되며, 무균 및 감염성 염증 반응에서 필수적 매개체로 작용합니다.
HMGB1 수준은 조직 손상, 감염, 패혈증, 관절염, 허혈-재관류 손상 등에서 증가하는 것으로 보고되었습니다.
