Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells.
CD40 regulates B cell development and maturation by inducing Ig isotype-switching and, with IL-4, protects B cells from surface Ig-induced apoptosis and promotes proliferation. Interaction of CD40 with CD154 (gp39) on T cells is crucial for T-B cell crosstalk and immune regulation.
5C3 is reported to activate B cells in in vitro functional assays.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.125 µg)/test.
A test is defined as the amount of antibody staining a cell sample in a final volume of 100 µL, with 10⁵–10⁸ cells/test (empirically determined).

Fluorochrome Information:
Super Bright 436 can be excited with a violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent.
Ensure the instrument is capable of detecting this fluorochrome.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD40 is a single-chain glycoprotein, member of the TNF/NGF superfamily, homologous to CD30.
Expressed on all B cells except plasma cells, and also found on epithelial cells, carcinomas, and lymphoid dendritic cells.
Essential for immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

AKNA transcription factor regulates CD40 expression for homotypic cell interactions.
Adaptor protein TNFR2 interacts with CD40 for signal transduction.
CD40-CD154 interaction is necessary for amyloid-beta-induced microglial activation, implicated in Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
