Product Specific Information
The monoclonal antibody ALY7 recognizes mouse LYVE-1, a transmembrane glycoprotein similar to CD44. Its extracellular domain contains a conserved hyaluronan binding domain. LYVE-1 is expressed on lymphatic and liver endothelial cells, as well as some macrophage populations.
The lymphatic system transports proteins and cells (especially dendritic cells) throughout the body, acting as immune surveyors. LYVE-1, along with podoplanin, PROX-1, Tie-2, and VEGFR-3, is a characteristic protein of lymphatic endothelial cells (LECs).
The ligand for LYVE-1 is hyaluronan, a large mucopolysaccharide. Although LYVE-1 can bind hyaluronan in vitro, the binding site in vivo is masked by sialylated O-linked glycan chains, requiring modification or unmasking for binding.
Applications Reported: The ALY7 antibody is used for immunohistochemical staining (IHC-F).
Applications Tested: Validated for IHC-F on fixed frozen mouse intestinal tissue at ≤1 µg/mL. Not validated for flow cytometry.
Filter Recommendation:
For eFluor™ 615 conjugate, use filters capturing 615 nm emission (e.g., Excitation 560/55, 585LP, Emission 645/75). Alexa Fluor™ 594 filter is acceptable.

Target Information
LYVE1 is a major receptor for hyaluronan (HA) on lymph vessel walls. It encodes a 322-residue type I integral membrane protein, sharing 41% similarity with CD44. The extracellular domain includes a single Link module, the HA-binding domain of the Link protein family.
Unlike CD44, LYVE1 colocalizes with HA on the luminal face of lymph vessels and is absent from blood vessels, making it a unique marker for lymphatic endothelium. LYVE-1 regulates leukocyte and tumor cell trafficking to lymph nodes and is expressed primarily on lymphatic vessel endothelium.
Diseases associated with LYVE1 dysfunction include Complete Androgen Insensitivity Syndrome.
