Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and is essential for brain development. Mutations, rearrangements, and amplifications in the ALK gene are associated with various tumors, including anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer.
Chromosomal rearrangements often create a fusion gene between ALK (chromosome 2) and NPM (chromosome 5), producing the oncogenic NPM-ALK fusion protein.
ALK encodes a receptor tyrosine kinase with transmembrane and extracellular domains, absent in the NPM-ALK fusion product. It shows high sequence similarity to LTK and plays a crucial role in neuronal development.
