Description:
This 5-329 monoclonal antibody recognizes human CD74 (MHC Class II-associated invariant chain), a 33–43 kDa nonpolymorphic type II membrane protein. CD74 is highly expressed on B cells, subsets of activated T cells, Langerhans cells, dendritic cells, and epithelial cells. It acts as a chaperone for MHC Class II (HLA-DR) proteins, aiding their transport to endocytic compartments during antigen presentation. CD74 interaction with CD44 and MIF is linked to NFκB and ERK activation. It also contributes to follicular B-cell maturation and marginal zone B-cell accumulation, and interacts with CXCR2 and CXCR4.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg)/test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test.

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB). The DRA gene consists of 5 exons encoding leader peptide, extracellular domains, transmembrane domain, and cytoplasmic tail. DRA lacks polymorphisms in the peptide-binding region and pairs with DRB1, DRB3, DRB4, and DRB5. DRB1 allelic polymorphisms determine peptide-binding specificity and are important in transplantation typing. HLA-DR is primarily expressed on antigen-presenting cells such as B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes. MHC class II molecules (DR, DQ, DP) present processed peptides to T-helper cells, initiating immune responses.
