Product Specific Information
eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell-surface protein with ecto-5'-nucleotidase activity.
Expression on myeloid cells (CD11b) is restricted to bone marrow. In humans, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in activation signaling.
CD73 is expressed on subsets of lymphocytes and increases during lymphocyte maturation. Memory CD4 T cells expressing CD73 resemble uncommitted primed precursor helper cells (Thpp) that can differentiate into Th1 or Th2 cells. CD73 is also found on regulatory T cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤ 0.125 µg/test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody titration recommended for optimal performance.

Target Information
CD73 (Ecto-5'-nucleotidase, 5'-ribonucleotide phosphohydrolase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides at neutral pH, with AMP as the preferred substrate.
It is a dimer of two 70 kDa subunits bound to the plasma membrane via glycosyl phosphatidyl inositol linkage. CD73 serves as a marker of lymphocyte differentiation; deficiency is associated with various immunodeficiency diseases.
Other 5'-nucleotidase forms exist in the cytoplasm and lysosomes, distinguished by substrate affinities and cofactor requirements.
The CD73 gene is localized to 6q14–q21. Defects may lead to joint and arterial calcification and intestinal tuberculosis. Two transcript variants encoding different isoforms are known.
