Product Specific Information

The RAM34 monoclonal antibody reacts with mouse CD34 (mucosialin). It is used to detect CD34⁺Sca-1⁺c-Kit⁺ cells. CD34 is a 90–120 kDa sialomucin family member expressed on capillary endothelial cells, bone marrow stroma, and a subset of mouse bone marrow cells. RAM34 enables purification of mouse hematopoietic stem cells (HSC) to near homogeneity.

CD34 on endothelial cells acts as a ligand for CD62L and plays a role in adhesion. Co-staining of mouse bone marrow cells with lineage markers (CD3, CD11b, Ly6G, TER-119, CD45R/B220) identifies RAM34⁺ cells with low or undetectable lineage marker expression.

Target Information

CD34 is a monomeric, highly glycosylated (111–115 kDa) surface protein expressed on various stem cell populations. It functions as a receptor involved in adhesion, differentiation, and proliferation of hematopoietic stem and progenitor cells. CD34 expression indicates a developmental state associated with altered adhesion and differentiation potential. It may serve as a scaffold for binding glycans and lectins, mediating stem cell attachment to the bone marrow matrix or stromal cells. The intracellular domain is phosphorylated by protein kinase C, suggesting a role in signal transduction.

Diseases associated with CD34 dysfunction include dermatofibrosarcoma and neurofibroma.
