Description:
The 4H11 monoclonal antibody reacts with human CD34 (mucosialin). CD34 belongs to a family including endoglycan and podocalyxin, which are transmembrane proteins with heavily glycosylated extracellular domains. CD34 was first identified on hematopoietic progenitors and is widely used as a marker for cells capable of engraftment. It is also expressed on mesenchymal stem cells, tumor cell lines, and vascular endothelium.
Epitopes of CD34 are classified as I, II, or III based on enzymatic cleavage sensitivity. The 4H11 antibody belongs to class III, reacting with a protein epitope.
Applications Reported:
Reported for use in flow cytometric analysis.
Applications Tested:
Pre-diluted and tested by flow cytometry on normal human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per test in a final volume of 100 µL. Applicable cell numbers: 10⁵–10⁸ cells/test.
Fluorochrome Information:
Alexa Fluor 700 emits at 723 nm and can be excited by the red laser (633–647 nm). Recommended optical setup: 685 LP mirror and 710/20 bandpass filter.

Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It functions as a stem cell marker, potentially involved in receptor-mediated endocytosis, adhesion, differentiation, and proliferation regulation. CD34 may mediate stem cell attachment to the bone marrow matrix or stromal cells and act as a scaffold for glycan attachment, facilitating lectin binding. The intracellular domain is phosphorylated by protein kinase C, suggesting a role in signal transduction.
