Target Information
Cyclin D1 (PRAD-1, bcl-1)는 세포 주기 조절의 핵심 단백질로, cdk4 및 cdk6과 결합하여 Rb 단백질을 인산화시킵니다. Cyclin D1은 여러 인체 종양(특히 mantle cell lymphoma)에서 과발현되는 원암유전자(proto-oncogene)로 알려져 있습니다. 또한 단백질 인산화, 유선 상피세포 증식, 지방세포 분화에 관여합니다.
Cyclin D1은 G1기 동안 합성되어 CDK4 또는 CDK6과 복합체를 형성하며, 성장 인자 자극 시 세포주기 G1/S 전이를 조절합니다. 성장 인자 자극이 없을 경우 Cyclin D1은 불안정하여 유비퀴틴 매개 분해를 통해 G1/S 정지에 관여합니다. Cyclin D1은 종양 억제 단백질 Rb와 상호작용하며, 이 유전자의 과발현이나 변이는 종양 형성에 기여할 수 있습니다.
