Target Information
Paraoxonase-1 (PON1) is a calcium-dependent ester hydrolase associated with apoA-I in HDL. PON1 prevents LDL oxidation and atherogenic oxidised-LDL formation. Human PON1 polymorphisms are linked to coronary artery disease. PON1 activity is lower in individuals prone to atherosclerosis, such as those with diabetes, hypercholesterolemia, or renal disease. PON2 is an intracellular protein protecting cells from oxidative damage. PON3 is similar to PON1 but differs in substrate specificity. Both PON1 and PON3 are implicated in reducing coronary artery disease risk. Human PON3 shares three conserved cysteine residues with PON1, suggesting their importance in vivo. PON1-based catalytic scavengers may serve as safe and effective agents against organophosphate intoxication.
