Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase activity and plays an important role in lymphocyte activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested for flow cytometric analysis of normal human peripheral blood cells. Use at 5 µL (0.8 µg)/test. A test is defined as the amount of antibody that stains a sample in 100 µL final volume. Cell number should range from 10⁵ to 10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample.

NovaFluor conjugates utilize Phiton technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover and spread.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, representing about 10% of lymphocyte surface proteins. It is absent in non-hematopoietic cells and tissues. CD45 plays a crucial role in lymphocyte development and antigen signaling, regulating Src-family kinases and JAK kinases, and modulating integrin-mediated signaling and apoptosis.
