Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed on all hematopoietic cells except circulating erythrocytes and platelets. The cytoplasmic portion exhibits tyrosine phosphatase activity, crucial for lymphocyte activation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells. Use 5 µL (0.8 µg)/test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails prior to combining with cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology with engineered nucleic acid dye structures designed to minimize spillover and spread.

Target Information

CD45 (LCA) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells and comprises ~10% of lymphocyte surface proteins. It is absent in non-hematopoietic tissues. CD45 regulates Src-family kinases, T- and B-cell receptor signaling, and cytokine receptor pathways. Isoforms arise from alternative splicing and vary among lymphoid subtypes. CD45 also modulates integrin-mediated signaling, apoptosis, and immune regulation.
